This discovery supports the use of fenbendazole as an complementary therapy to chemotherapy and radiotherapy as well as metabolic therapies. Fenbendazole is an approved anthelmintic in the US for beef and dairy cattle including lactating dairy cattle, swine, turkeys, equine, dogs, goats, hoofed wildlife and zoo animals. The first notable observation of an anti-brain tumor activity of fenbendazole occurred when an animal facility at John Hopkins University School of Medicine in Baltimore, Maryland started providing fenbendazole containing food to the mice colony to eliminate pinworm infections. During the feeding period, one of the orthotopic brain tumor models stopped forming tumors, whereas before the fenbendazole therapy, this model provided consistent, reproducible tumor engraftment. This prompted an investigation of commercially available benzimidazoles: fenbendazole, thiabendazole, mebendazole and albendazole, by first using mebendazole and albendazole since they were the only FDA approved benzimidazole products for human use (also for parasite control).

The mode of action of the benzimidazole family anthelmintics against parasites was that these products can exert cellular toxicity by binding to tubulin molecules and thereby disrupting their polymerization and microtubule formation in essence starving the cell to death by preventing glucose uptake and subsequently killing the parasite. In numerous studies, this family of products has been shown to have no adverse effect on mammalian cells except when the cell undergoes rapid development as is the case with tumor development. It appears through several studies that the benzimidazoles seem to target cancer cells preferentially over normal cells. These studies thus far have shown that in vivo studies against cancers, including non-small cell lung cancer, adrenocortical carcinoma, lymphoma, leukemia, colorectal cancer and now glioblastoma multiforme (GMB –the most common and aggressive brain cancer in humans).
