Inhibition of SARS-CoV 3CL protease by flavonoids

Intriguingly, some flavonoids also have antiviral activity17. Specifically, apigenin, luteolin, quercetin, amentoflavone28, quercetin, daidzein, puerarin, epigallocatechin, epigallocatechin gallate, gallocatechin gallate29 and kaempferol30 were reported to inhibit the proteolytic activity of SARS-CoV 3CLpro. Therefore, the antiviral effect is presumed to be directly linked to suppress the activity of SARS-CoV 3CLpro in some cases. However, the systematic analysis using various scaffolds of flavonoids was not reported. In order to find the best scaffold to inhibit the function of SARS-CoV 3CLpro, an assay with various flavonoid derivatives classified in 10 scaffolds was built and performed. Among them, the best compounds were one flavonol, herbacetin and two flavones, rhoifolin and pectolinarin.

The presence of additional 8-hydroxyl group of herbacetin was expected to be critical to acquire its high binding affinity around the S1 and S2 sites. The occupation of the S1 and S2 sites by carbohydrate groups of rhoifolin and pectolinarin was anticipated to be another way of getting high affinity to SARS-CoV 3CLpro in glycosylated flavonoids.

Hey guys, I am finally done with the whole Covid thing by now. I still have slight hypoxia, but it is very doable.

So adding back to my previous post:

1. Quinine (even from tonic water, but make sure it really has it, some brands in my country do not) does help, but I do believe it helps prevent the replication of the virus. In any case, it should be done very early and then it should theoretically stop the onset.

2. Since one of the mechanisms that the virus causes hypoxia with is by disrupting the porphyrin cycle, making the iron that would normally carry oxygen loose in the body and having it cause oxidative damage, it would then make sense to have antioxidants. This thread has plenty of ideas for that. And you need good iron and zinc intake to make new red blood cells.


So in conclusion, my ideal scenario for treatment of this virus would be:

1. Prevention, if that fails, attempting to stop the virus from replicating.

2. Mitigate the damage of loose oxidizing iron in your body

3. Make new red blood cells that will be healthy and be able to perform their function so that you recover from being hypoxic

All in all, not giving health advice, obviously, just speaking from experience.

Fantastic thread! I’m glad it was pinned or I wouldn’t have seen it! I can’t wait to read through it all but I want to share my own experience. Not covid, but I find it logically similar.

I used to get incredibly bad colds that were getting worse every year. I would be completely out of it miserable for a month or more. It finally hit me that it felt like a similar reaction that I had with seasonal allergies. So I stopped treating a cold. Instead, I treated it like a respiratory bug, that created an allergic reaction. And it’s a cure. Now, as soon as I feel it coming on, I kill the germs by inhaling diffused oregano as much as I can. I’ll inhale the mist straight from my diffuser as close and as much as I can stand to get the oregano into my respiratory tract as much as possible. And I treat the allergic reaction to it with eucalyptus and camphor. It’s taken it from being a three or four week ordeal of misery and turned it into just a few days. If I can catch it early enough, I don’t even get miserable at all. Just a few days of being mildly stuffy.

So say what you will. Covid, at the base functionality of it sounds to me like my really bad colds. Certainly, it can get worse. But it’s a respiratory illness with an allergic reaction. Kill the germs with oregano before they take hold and create all the other problems. Manage whatever allergic reaction that happens. And now I know all the things about high dose vitamin c. And I definitely won’t take anything to suppress a fever either. That’s the body’s natural defense to kill germs. Let it.
