Detecting Airborne Ebola Virus

Release Date:
April 20, 2021
Natural outbreaks of the Ebola virus, while severe, are typically isolated and usually affect no more than a few hundred people at a time. However, from 2014-2016, infections from this deadly virus caused more than 11,000 deaths in West Africa. During this time, several cases of Ebola virus disease were also diagnosed in other countries, including the United States, due to infected travelers from West Africa that had unknowingly harbored and incubated the virus while en-route to their respective destinations.

By the time a person infected with Ebola virus becomes symptomatic, they are typically starting to shed the virus. During an outbreak, health care workers and family members of patients are often the first to respond and provide care. In this role, they are at a high risk of becoming infected with the virus as well. Therefore, it is important to understand how we can best prevent transmission in both clinical and home settings.

Human to human transmission of the Ebola virus occurs primarily through direct contact and exposure to the blood or other bodily fluids of infected patients. However, there have been new infections that occurred without documented contact between a patient and health care provider or family member. While these cases are uncommon, it is possible that a small fraction of the cases of Ebola virus infection may be the result of exposure to small droplets or aerosols containing the virus.

Laboratory studies have shown that the Ebola virus can remain infectious outside of the body for long periods of time. The virus can survive in blood samples on various surfaces for several days, even in hot and humid conditions that would typically kill most other viruses and bacteria. In an aerosolized form, the Ebola virus can survive for over an hour. Additionally, laboratory experiments have demonstrated that inhaling small amounts of Ebola virus can be fatal [1], and there are examples of Ebola virus disease being transmitted between individuals in close proximity, even though they had never been in direct contact with each other [2].

However, assessing the risk of transmission via droplets or aerosols is a complicated process. While the minimum amount of virus required to cause infection is thought to be very low, so far it has not been possible to determine a definitive value. This is because commonly used testing methods are often not sensitive enough to detect or measure the amount of infectious virus in the air. To address these challenges, researchers at the Department of Homeland Security (DHS) Science and Technology Directorate’s (S&T) National Biodefense Analysis and Countermeasures Center (NBACC) designed and conducted a study to optimize methods for collecting and measuring very small amounts of Ebola virus in the air.

“Ebola can be both a national security and public health concern,” explained Lloyd Hough, who leads S&T’s Hazard Awareness and Characterization Technology Center. “We’re looking forward to applying these methods to better characterize the risks associated with Ebola virus, and are hopeful that others can benefit from these techniques as well.”

NBACC researchers assessed and compared multiple devices designed for collecting microorganisms from the air. They concluded that filters made of gelatin were the best for collecting infectious Ebola virus from the air and were also the easiest and safest type of sampling device to use. The researchers utilized a cell line, developed by the Centers for Disease Control and Prevention, in an assay designed to measure the amount of infectious Ebola virus present in a sample. The cell line glows when infected by the virus, which enabled the researchers to differentiate Ebola virus infection from other causes of cell death.

After testing various sampling and assay methodologies, the NBACC researchers found that the combination of the gelatin filter samplers and improved assay was easier to use, more reliable, and nearly ten times more sensitive than the previous methods utilized to measure the amount of infectious Ebola virus in air samples. The results of these experiments have been published in two peer-reviewed journal articles [3,4], which will enable researchers at other institutions to understand and utilize these newly developed sampling and assay methodologies.

“Our ability to detect Ebola virus in air samples at levels that are ten-fold lower than what was possible with previous methods, will enable us to provide a better understanding of the aerosol hazard posed by this virus,” said Mike Schuit, an investigator in the Aerobiology group at NBACC.

The new sampling and assay methodologies developed by NBACC researchers, as well as the data from associated studies, will be useful in a number of ways. For example, these methods are currently being employed in a study in partnership with the National Institute of Allergy and Infectious Diseases (NIAID) to determine the minimum amount of Ebola virus that needs to be inhaled in order to cause infection or death in an animal model of Ebola virus disease. This study will also determine whether infected animals produce aerosols containing Ebola virus when they breathe, which will help scientists to better understand the potential for natural aerosol transmission of the virus. Various DHS components and partners, along with the Department of Defense and the Department of Health and Human Services, will use these data to conduct hazard modeling and gain a better understanding of the potential risk for airborne transmission of Ebola virus and how it may impact health workers.

Taking lessons learned from the response to Ebola virus, the NBACC researchers are conducting similar studies with SARS-CoV-2, the virus that causes COVID-19. This includes studying the performance of aerosol samplers with SARS-CoV-2, and optimization of methodologies to detect small quantities of SARS-CoV-2 in the air. Furthermore, the NIAID partnership has been extended to examine how much virus it actually takes to start a new infection when aerosol particles containing SARS-CoV-2 are inhaled. As with the Ebola study, this study will also measure whether infectious virus is present in the exhaled breath of infected animals to better understand how COVID-19 spreads in human populations and inform strategies to prevent its continued spread.

In all practicality, the only way to make the Ebola Virus 'airborne' is by artificial manipulation of its genome. In other words, turning it into a bioweapon.

For EBV to mutate to that degree would make it far less virulent.

I would trust nothing at this point, wear a mask for what its worth, your screwed if you don't have your own well. Maybe only drink bottled water and grow and raise your own food.

We all know what lock down is like guess those that are aware maybe do your own lock down longer.

If they spray or aerosol dispersed nothing we can do, unless you got some kind of wicked air purifier but still your food outside will have it on it.

I don't know, but nothing would surprise me at this rate the world is going.

And I would like to add:

I don't trust tptb, but my first thoughts at the start of this pandemic was how come they keep asking us to stay inside. I told my husband I think we are at war. No one was even allow to be out at campgrounds, no fires etc. Not even boat launches which made no sense except for don't go outside.

Just last week provincial Doug ford in Ontario Canada here again said Don't go outside only for essentials.

Makes you wonder.....

Symptoms may appear anywhere from 2 to 21 days after contact with the virus, with an average of 8 to 10 days. The course of the illness typically progresses from “dry” symptoms initially (such as fever, aches and pains, and fatigue), and then progresses to “wet” symptoms (such as diarrhea and vomiting) as the person becomes sicker.

Primary signs and symptoms of Ebola often include some or several of the following:
•Fever
•Aches and pains, such as severe headache and muscle and joint pain
•Weakness and fatigue
•Sore throat
•Loss of appetite
•Gastrointestinal symptoms including abdominal pain, diarrhea, and vomiting
•Unexplained hemorrhaging, bleeding or bruising

Other symptoms may include red eyes, skin rash, and hiccups (late-stage).

Many common illnesses can have the same symptoms as EVD, including influenza (flu), malaria, or typhoid fever.

EVD is a rare but severe and often deadly disease. Recovery from EVD depends on good supportive clinical care and the patient’s immune response. Studies show that survivors of Ebola virus infection have antibodies (proteins made by the immune system that identify and neutralize invading viruses) that can be detected in the blood up to 10 years after recovery. Survivors are thought to have some protective immunity to the type of Ebola that sickened them.

Johnson & Johnson Joins World Health Organization

Prevent Spread of Ebola in West Africa

Up to 200,000 Johnson & Johnson Ebola vaccine regimens will be made available as part of a WHO early access clinical program now underway in Sierra Leone

Company’s Ebola vaccine regimen also receives Prequalification from the WHO

The Johnson & Johnson vaccine regimen is designed to be used proactively to induce immunity against Ebola in adults and children
NEW BRUNSWICK, N.J., May 13, 2021 – Johnson & Johnson (NYSE: JNJ) (the Company) today announced the World Health Organization (WHO) and the government of Sierra Leone have begun administering the Company’s Ebola vaccine regimen as part of a WHO early access clinical program aimed at preventing further spread of Ebola in West Africa. The vaccine regimen, developed by the Janssen Pharmaceutical Companies of Johnson & Johnson (Janssen) in collaboration with Bavarian Nordic A/S, is being donated to the WHO by Janssen for the purposes of the early access clinical program. Johnson & Johnson also announced that its Ebola vaccine regimen has received Prequalification from the WHO, which will help accelerate its registration in countries where Ebola is a persistent public health threat and facilitate broader access to people at risk of exposure to this virus.

Health authorities in Guinea officially declared a new Ebola outbreak in February 2021 after the West African country experienced its first cases of disease since the end of the 2014-2016 Ebola outbreak – which was the worst on record. Preliminary data obtained through genetic sequencing suggest that the new outbreak was caused by the same virus strain also responsible for the 2014-2016 outbreak and was likely reintroduced by a survivor. The continuation of the outbreak, which has caused at least 23 cases and 12 deaths, and the persistence of Ebola virus in human and animal hosts underscores the importance of proactive vaccination efforts to prevent the further spread of the virus and to be prepared for potential new outbreaks in the future.

“Johnson & Johnson’s vision is to help prevent Ebola outbreaks before they start. WHO Prequalification of our vaccine regimen and the deployment to West Africa are important steps forward in reaching this goal and an important milestone for epidemic preparedness,” said Paul Stoffels, M.D., Vice Chairman of the Executive Committee and Chief Scientific Officer of Johnson & Johnson. “By working closely with the WHO and national governments, we will be able to quickly support efforts to protect populations at risk and help end this latest outbreak. This proactive approach is essential if we are to solve the growing threat of infectious diseases, including Ebola.”

Johnson & Johnson is grateful to its global strategic partners who have helped to support and co-fund the Ebola vaccine regimen, including Bavarian Nordic A/S, the Biomedical Advanced Research and Development Authority (BARDA) and the National Institutes of Health (NIH) at the U.S. Department of Health and Human Services (HHS), and the Innovative Medicines Initiative (IMI) funded through the EU Horizon 2020 program.

WHO Early Access Clinical Program
Administration of the Johnson & Johnson Ebola vaccine regimen in West Africa will be directed by the WHO and national governments in accordance with the protocol for the early access clinical program, which incorporates interim 2019 recommendations from the WHO’s Strategic Advisory Group of Experts on Immunization (SAGE). The goal of the initiative is to establish a protective geographic barrier against the spread of Ebola beyond Guinea, beginning in the neighboring nation of Sierra Leone. The program administered its first doses of the Johnson & Johnson Ebola vaccine regimen this week in Sierra Leone’s Kambia District, and is expected to expand to additional countries in the region as local preparations are completed.

The early access clinical program plans to administer the vaccine to up to 200,000 individuals beginning with health workers, other frontline workers and others at increased risk of exposure to the Ebola virus (adults and adolescents 14 years and above on a case-by-case basis or depending on national regulations). Pregnant women and HIV-positive adults who are otherwise healthy are permitted to enroll in the program.

Sierra Leone has prior experience administering the Johnson & Johnson Ebola vaccine. In 2015, during the West African epidemic, the country’s Ministry of Health and the College of Medicine and Allied Health Sciences (COMAHS) at the University of Sierra Leone collaborated with Johnson & Johnson on the first clinical study of the vaccine in an Ebola-affected country (“EBOVAC-Salone”), which took place in Sierra Leone’s Kambia district.

“We are moving with urgency and purpose to bring all of our available resources to help prevent the spread of this latest Ebola outbreak in Guinea,” said Ruxandra Draghia-Akli, M.D., Ph.D., Global Head, Global Public Health Research & Development, Johnson & Johnson. “We believe that, through the preventive use of Ebola vaccines, the global health community can help protect vulnerable communities living under the threat of this disease.”

WHO Prequalification
The Johnson & Johnson Ebola vaccine regimen, Zabdeno® (Ad26.ZEBOV) and Mvabea® (MVA-BN-Filo), was granted WHO Prequalification in April 2021. This follows Marketing Authorisation for the vaccine which was granted by the European Commission in July 2020 for the active immunization for the prevention of Ebola Virus Disease caused by the Zaire ebolavirus species in individuals aged one year and above. Both WHO Prequalification and the European Commission authorization were supported by a multi-country clinical program, including 15 clinical studies sponsored by Janssen across three continents. Discussions are ongoing with the U.S. Food and Drug Administration regarding the approval of the vaccine regimen in the U.S.

WHO Prequalification is often a prerequisite for national registrations of new vaccines and medicines in developing countries. Johnson & Johnson now looks forward to collaborating with the WHO’s African Vaccine Regulatory Forum (AVAREF) to progress national registrations of the Company’s Ebola vaccine regimen. The Company’s Ebola vaccine regimen is designed to be used proactively to induce immunity against Ebola virus disease in adults and children.

Johnson & Johnson’s Commitment to Ebola & Pandemic Preparedness
Johnson & Johnson is one of the few innovative healthcare companies in the world today that is actively advancing science across multiple disease areas with the aim of strengthening public health.

We accelerated the development of our Ebola vaccine regimen in 2014 in response to the worst Ebola outbreak on record, which took place in West Africa from 2014-2016 and caused more than 11,000 deaths. In 2019, in response to the second-worst outbreak which took place 2018-2020 in the Democratic Republic of the Congo (DRC), Johnson & Johnson announced it would provide its Ebola vaccine regimen to assist immunization efforts in the affected region and in neighboring Rwanda through the UMURINZI vaccination campaign. This marked the first widespread deployment of Ebola vaccines in an outbreak setting.

The Rwandan Ministry of Health continues to administer the vaccine regimen through the UMURINZI campaign, which recently achieved the milestone of reaching 200,000 individuals who are at risk of exposure to Ebola. To date, more than 225,000 individuals participating in clinical trials and vaccination initiatives have received at least the first dose of the Johnson & Johnson Ebola vaccine regimen, including 180,000 who have been fully vaccinated.

Johnson & Johnson Ebola Vaccine Regimen
The European Commission-approved Johnson & Johnson preventive Ebola vaccine regimen, Zabdeno® (Ad26.ZEBOV) and Mvabea® (MVA-BN-Filo), utilizes a non-replicating viral vector strategy in which viruses – in this case adenovirus serotype 26 (Ad26) and Modified Vaccinia Virus Ankara (MVA) – are genetically modified so that they cannot replicate in human cells. In addition, these vectors carry the genetic code of several Ebola virus proteins in order to trigger an immune response. The Ebola vaccine regimen was developed and is manufactured using Janssen’s proprietary AdVac® technology.

Johnson & Johnson also acknowledges its many strategic partners in the ongoing global clinical program for the vaccine regimen, including Bavarian Nordic A/S, Centre Muraz, Coalition for Epidemic Preparedness Innovations (CEPI), College of Medicine and Allied Health Sciences (COMAHS, University of Sierra Leone), Democratic Republic of the Congo Ministry of Public Health, Republic of Rwanda Ministry of Health and Rwanda Biomedical Center, Emory University’s Project San Francisco (Kigali) / Center for Family Health Research, Emory University, Epicentre, Grameen Foundation, Inserm, Inserm Transfert, Institut National de Recherce Biomédicale (INRB), London School of Hygiene & Tropical Medicine (LSHTM), Médecins Sans Frontières (MSF), Rinda Ubuzima, Sierra Leone Ministry of Health and Sanitation, Uganda Virus Research Institute (UVRI), Université de Kinshasa (UNIKIN), University of Antwerp, University of Oxford, Walter Reed Army Institute of Research (WRAIR), World Health Organization, World Vision Ireland, Wellcome Trust, Vibalogics, and all the people who have participated in the Ebola vaccine clinical trials.

Ebola vaccination starts in Sierra Leone to prevent new outbreak

Preventive Ebola vaccination will continue over next few days in several districts sharing borders with Guinea

Ebola vaccination starts in Sierra Leone to prevent new outbreak

ANKARA

Sierra Leone has launched the Ebola vaccination campaign to prevent a new outbreak in the West African country, according to a statement.

The campaign kicked off on Wednesday as the country takes proactive public health measures to protect people at high risk of infection in case of any potential cross-border transmission of the disease from the ongoing outbreak in neighboring Guinea.

The preventive Ebola vaccination will continue over the next few days in several districts sharing borders with Guinea, according to the World Health Organization (WHO).

About 16,000 people have been targeted for vaccination in the selected border districts, and they will receive two doses of the vaccine given approximately eight weeks apart.

“This strong partnership, led by the Ministry of Health and Sanitation is a strategic step towards protecting the health of the population in Sierra Leone and a clear demonstration of the concern and care for the frontline workers who may be at risk,” Steven Velabo Shongwe, WHO representative in Sierra Leone, said in a statement on Wednesday.

Last week, the Democratic Republic of Congo (DRC) announced the end of the 12th Ebola outbreak -- three months after the first case was reported in the North Kivu province.

The disease caused global alarm in 2014, killing more than 11,300 people and infecting an estimated 28,600 as it swept through Guinea, Liberia, and Sierra Leone in the course of the epidemic.

Ebola, a tropical fever that first appeared in 1976 in Sudan and the DRC, is transmitted to humans from wild animals.

Currently, there is an ongoing Ebola outbreak in the West African country of Guinea, which began in February.
