However, Nicotine is a bioactive compound in cigarettes that exerts rewarding effects by activating nicotinic acetylcholine receptors (nAChRs) in the central nervous system. Repetitive nicotine intake modifies plasticity in the central nervous system, leading to nicotine dependence [2].

Nicotinic acetylcholine receptors, or nAChRs, are receptor polypeptides that respond to the neurotransmitter acetylcholine. ... At the neuromuscular junction they are the primary receptor in muscle for motor nerve-muscle communication that controls muscle contraction.

In particular, the α7-nAChR is emerging as an important player in the HIV field, since it is expressed not only in the brain, but also in a wide variety of immune cells that are targeted during HIV infection, such as macrophages, monocytes, B-lymphocytes, and T-lymphocytes (CD4+) (Wang et al. 2003; van der Zanden et al. 2012; Kawashima et al. 2015), making it a suitable target for treatment development.

Due to the presence of α7-nAChR on both neuronal and immune cells, and its known participation in HIV settings, this review will focus on the role of α7-nAChRs in two different but equally important aspects of HIV: inflammation (neuroinflammation and peripheral inflammation) and HAND, both of which severely compromise the quality of life of patients infected with HIV. Here, we review the role of α7-nAChRs in these HIV-derived consequences and suggest the receptor as a possible pharmacological target to treat/ameliorate both HAND and inflammation.

Altogether, these studies suggested a role for nAChRs in HIV. However, considering the pharmacological properties and different roles of the α7-nAChR, a closer look is justified. Below we describe the proposed functions/roles of this receptor to then highlight studies specifically relating to HIV-induced inflammation and HAND, where α7-nAChRs play a critical part.

In addition to its central role in the CNS, the α7-nAChR is also present in immune cells. As a result, several efforts have been made to elucidate its role in these cells. It is now clear that the α7-nAChR is an essential regulator of inflammation in macrophages through the CAP (Tracey 2007). The anti-inflammatory activity of the α7-nAChR occurs by the ACh-induced activation of the receptor to inhibit the production of proinflammatory cytokines in macrophages, thereby modulating immune responses and the progression of inflammatory diseases, avoiding organ and systemic damage (Tracey 2009). Altogether, these studies highlight the importance of this neuroimmune tract for the well-being of healthy people and accents the α7-nAChR’s pivotal role in the innate immune response necessary to resolve inflammation.

While SARS-CoV-2 uses angiotensin converting enzyme 2 (ACE2) as the receptor for cell entry, it is important to examine other potential interactions between the virus and other cell receptors. Based on the clinical observation of low prevalence of smoking among hospitalized COVID-19 patients, we examined and identified a "toxin-like" amino acid (aa) sequence in the Receptor Binding Domain of the Spike Glycoprotein of SARS-CoV-2 (aa 375-390), which is homologous to a sequence of the Neurotoxin homolog NL1, one of the many snake venom toxins that are known to interact with nicotinic acetylcholine receptors (nAChRs). We present the 3D structural location of this "toxin-like" sequence on the Spike Glycoprotein and the superposition of the modelled structure of the Neurotoxin homolog NL1 and the SARS-CoV-2 Spike Glycoprotein. We also performed computational molecular modelling and docking experiments using 3D structures of the SARS-CoV-2 Spike Glycoprotein and the extracellular domain of the nAChR α9 subunit. We identified a main interaction between the aa 381-386 of the SARS-CoV-2 Spike Glycoprotein and the aa 189-192 of the extracellular domain of the nAChR α9 subunit, a region which forms the core of the "toxin-binding site" of the nAChRs. The mode of interaction is very similar to the interaction between the α9 nAChR and α-bungarotoxin. A similar interaction was observed between the pentameric α7 AChR chimera and SARS-CoV-2 Spike Glycoprotein. The findings raise the possibility that SARS-CoV-2 may interact with nAChRs, supporting the hypothesis of dysregulation of the nicotinic cholinergic system being implicated in the pathophysiology of COVID-19. Nicotine and other nicotinic cholinergic agonists may protect nAChRs and thus have therapeutic value in COVID-19 patients.

The ingredients of e-liquids and the component parts of vaporizers are all well known, all of the ingredients in e-liquids are food grade and likely FDA approved, even if the finalized combination product is not.

Nicotine on its own is no more harmful than caffeine.

Though, it is weird how they conflated nicotine vaping with the illegal marijuana vaping that was causing EVALI due to tainted THC cartridges which contained Vitamin E Acetate. And it is also strange how quickly EVALI disappeared as COVID appeared.
