In a rage, something Baldwin is infamous for, Alec obviously loaded the pistol himself and murdered the woman in retaliation for the camera crew walkout only a few hours earlier.

Alec Baldwin was handed a pistol that had been confirmed to be empty by the crew.

A few minutes later while pretending to be practicing his fast draw in the church, he used that pistol to shoot and kill the woman who had just argued in support of her camera crew who eventually walked off the set due to poor working conditions and not getting paid by skinflint Baldwin.
