Role of Ionizable Lipids in SARS-CoV-2 Vaccines As Revealed by Molecular Dynamics Simulations: From Membrane Structure to Interaction with mRNA Fragments:

“ Self-Assembly of Lipid Mixtures. To simulate the experimental vaccine mixing step, we studied the self-assembly of lipid mixtures containing charged ILs with and without the presence of an RNA fragment. During the self-assembly process, we observed four major possible system organizations: (a) intact bilayer, (b) porous bilayer, (c) cubic phase, and (d) nanodisks (Figure Figure33). The intact bilayer is a lamellar phase, i.e., a two-dimensional (2D) structure with no disturbances in the bilayer plane and no contact between individual bilayers in the z-direction (except for interactions involving PEG chains). The porous bilayer is also separated in the z-direction but contains pores in the bilayer plane. The cubic phase is interconnected in all three dimensions (3D), while nanodisks are spheroidal lipid particles surrounded by water molecules. The final structure depends on the system’s composition, size, and level of hydration, but generally we observed that ILs-containing mixtures preferred various nonlamellar phases.”
