Potential COVID-19 Achilles Heel Discovered: MicroRNA Molecules May Repress the Replication of Human Coronaviruses


Smart virus outfoxed: Russian researchers have found MicroRNA molecules potentially capable of repressing the replication of human coronaviruses.


HSE University researchers have found microRNA molecules that are potentially capable of repressing the replication of human coronaviruses, including SARS-CoV-2. It turns out that the virus uses miRNA hsa-miR-21-3p to inhibit growth in the first stages of infection in order to delay the active immune response. The results of the research will be published today (September 14, 2020) in the journal PeerJ.

After the virus gets inside the cell, it starts actively interacting with various in-cell molecules. One such molecule class is microRNAs (miRNAs), which are small RNAs whose main function is to regulate gene expression. When a virus enters, miRNAs start binding certain parts of its genome RNA, which leads to the destruction of virus RNAs. Such an attack can stop virus replication completely. However, in cases when miRNAs are not very “aggressive,” such interactions do not destroy the virus but rather slow down its replication. This scenario is beneficial for the virus since it helps avoid a fast immune response in the cell. And some of the viruses purposefully accumulate host miRNA binding sites. This becomes their advantage: viruses with more binding sites survive and reproduce better, which leads to their evolutionary domination.

HSE University researchers have become the first in the world to discover genetic predisposition to severe COVID-19. The results of the study were published in the journal Frontiers in Immunology.

T-cell immunity is one of the key mechanisms used by the human body to fight virus infections. The staging ground for cell immunity development is the presentation of virus peptides on the surface of infected cells. This is followed by activation of T lymphocytes, which start to kill the infected cells. The ability to successfully present virus peptides is largely determined by genetics. In human cells, human leukocyte antigen class I (HLA-I) molecules are responsible for this presentation. The set of six such molecules is unique in every human and is inherited from an individual’s parents. In simple terms, if the set of alleles detects the virus well, then the immune cells will detect and destroy the infected cells fast; if a person has a set that is bad at such detection, a more severe case of disease is more likely to be observed.

Now a research team from La Jolla Institute for Immunology (LJI), The University of Liverpool and the University of Southampton has uncovered an interesting clue. Their new study suggests that people with severe COVID-19 cases may be left with more of the protective “memory” T cells needed to fight reinfection.

“The data from this study suggest people with severe COVID-19 cases may have stronger long-term immunity,” says study co-leader LJI Professor Pandurangan Vijayanand, M.D., Ph.D.

Research Shows Exposure to Common Cold Coronaviruses Can Teach the Immune System to Recognize SARS-CoV-2

Your immune system’s “memory” T cells keep track of the viruses they have seen before. This immune cell memory gives the cells a headstart in recognizing and fighting off repeat invaders.

Now, a new study led by scientists at La Jolla Institute for Immunology (LJI) shows that memory helper T cells that recognize common cold coronaviruses also recognize matching sites on SARS-CoV-2, the virus that causes COVID-19.

The research, published on August 4, 2020, in Science, may explain why some people have milder COVID-19 cases than others — though the researchers emphasize that this is speculation and much more data is needed.

“We have now proven that, in some people, pre-existing T cell memory against common cold coronaviruses can cross-recognize SARS-CoV-2, down to the exact molecular structures,” says LJI Research Assistant Professor Daniela Weiskopf, Ph.D., who co-led the new study with LJI Professor Alessandro Sette, Dr. Biol. Sci. “This could help explain why some people show milder symptoms of disease while others get severely sick.”

New Research Finds Most People Are Naturally Armed Against the COVID-19 Coronavirus

The majority of the population can produce neutralizing antibodies against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in severe cases of coronavirus disease 2019 (COVID-19), according to a study published today (February 11, 2021) in the open-access journal PLOS Pathogens by Michael Mor of Tel Aviv University, and colleagues. Moreover, the results support the use of combination antibody therapy to prevent and treat COVID-19.

The COVID-19 pandemic, caused by SARS-CoV-2, has had a profound impact on global public health. Neutralizing antibodies that specifically target the receptor-binding domain (RBD) of the SARS-CoV-2 spike protein are thought to be essential for controlling the virus. RBD-specific neutralizing antibodies have been detected in convalescent patients – those who have recovered from COVID-19. Some of the recoverees tend to have robust and long-lasting immunity, while others display a waning of their neutralizing antibodies. The factors associated with an effective, durable antibody response are still unclear.

New York Finds Its Own Covid Variants. The News Is Not Good


In a hostile takeover of sorts, SARS-CoV-2 variants have emerged the world over, threatening progress made towards ending the Covid-19 pandemic. Certain mutations confer some variants the ability to evade vaccines, whereas others make the virus far more transmissible and more deadly. Here, we analyze the genetic makeup of another one of these shifty variants identified in New York (B.1.526). This strain, in particular, carries several well know mutations known to be troublesome and some of its very own.

The variant was first identified in late November by researchers at the California Institute of Technology Biology and Biological Engineering Division grew to account for nearly 30% of genomes sequenced from New York through mid-February 2021, as shown in the table below.

Fury at ‘do not resuscitate’ notices given to Covid patients with learning disabilities

NHS figures released last week show that in the five weeks since the third lockdown began, Covid-19 accounted for 65% of deaths of people with learning disabilities. Figures from the Office for National Statistics show that the rate for the general population was 39%, although the two statistics are drawn from different measurements.

People with learning disabilities have been given do not resuscitate orders during the second wave of the pandemic, in spite of widespread condemnation of the practice last year and an urgent investigation by the care watchdog.

Mencap said it had received reports in January from people with learning disabilities that they had been told they would not be resuscitated if they were taken ill with Covid-19.

There is yet more good news on vaccine effectiveness. A study based on nearly 9000 coronavirus tests done on healthcare workers in Cambridge, UK, has found that asymptomatic infections fell by 75 per cent 12 days after they got one dose of the Pfizer/BioNTech vaccine. There was a similar reduction in symptomatic infections.

Review into “vaccine passports” launched in UK

The UK government has begun a review of whether to introduce certificates showing someone has had a covid-19 vaccine. So-called vaccine passports could be used for international travel and for allowing people to enter places like theatres or pubs, Prime Minister Boris Johnson said today.

“There are deep and complex issues that we need to explore, about what the role is for government in mandating people to have such a thing or, indeed, banning people from doing such a thing,” Johnson said. “We can’t be discriminatory against people who, for whatever reason, can’t have the vaccine.” Led by cabinet office minister Michael Gove, the review’s findings are due before England’s fourth stage of lockdown easing, on 21 June.

Iceland has said they will, in future, let people with a certificate of covid-19 vaccination enter without quarantining or taking a coronavirus test, and Greece, whose economy is dependent on tourism, is also pushing the European Union to press ahead with organising vaccine passports for this summer.

Within the UK, some firms have said they plan to insist on new employees having been vaccinated. But there are still many unknowns about the effects of vaccination, including how much it cuts transmission, and how long immunity lasts for.

Vaccine passports “are feasible, but not all the pieces are in place yet,” says Melinda Mills at the University of Oxford, co-author of a Royal Society report that sets out 12 criteria that need to be satisfied before they can be used.

New Hampshire opened up pretty much fully in August and has stayed open this whole time.

Honestly I couldn't tell you what restrictions we have aside from a mask mandate, restaurants have been able to do full capacity since August I'm not aware of anything that really ever shut down.

Schools are hit or miss, some areas doing in-person learning and some remote... they've bounced back and forth depending on case load.

People around here for the most part take the virus seriously, and I think that goes way further than any government edict.

We started allowing tourism in July, everyone that normally goes to Vermont and Maine comes here now... hasn't been an issue.

The vaccines work in a similar fashion. They hijack human cells from preforming their God given natural duties and force them to build neurotoxic gain of function spike proteins with prion like domains which neither protect you from getting infected or from infecting others.
