A nicotinic hypothesis for Covid-19 with preventive and therapeutic implications

Nicotine may be suggested as a potential preventive agent against Covid-19 infection. Both the epidemiological/clinical evidence and the in silico findings may suggest that Covid-19 infection is a nAChR disease that could be prevented and may be controlled by nicotine. Nicotine would then sterically or allosterically compete with the SARS-CoV-2 binding to the nAChR. This legitimates the use of nicotine as a protective agent against
SARS-CoV-2 infection and the subsequent deficits it causes in the CNS. Thus, in order to prevent the infection and the retro-propagation of the virus through the CNS, we plan a therapeutic assay against Covid-19 with nicotine (and other nicotinic agents) patches or other delivery methods (like sniffing/chewing) in hospitalized patients and in the general population.



COMMENTS:
Paper above regarding the binding of S1 Spike to acetylcholine receptors.
- Some symptoms are neurological - and it is becoming extremely obvious that this is the main route of attack of the V upon the nervous system i.e via the nACh receptors.
- This paper provides some fascinating observations regarding the effect of nicotine.
- It also identifies the exact sequence of amino-acids within the Spike that are responsible for the binding to nACh receptors.
- This is the sequence - YQTQTNSPRRAR
- This sequence is very similar to sequences from snake neuro-toxins and from Rabies sequences that also operate by binding strongly to acetylcholine receptors, and consequently are also highly neuro-toxic.
- It seems that the part of the S1 Spike that has affinity for the acetylcholine (nACh) receptors is the Furin Cleavage site PRRA, the very site which is missing from all other Human beta-coronaviruses - hence most likely to have been added by gain-of-function engineering (lab-origin).
- Based on these observations, it would seem that nicotine, by binding to acetylcholine receptors in preference to the Spike, will greatly reduce the incidence of neurological pathologies following infection or even possibly vaccination, too!.
- Interestingly, Ivermectin is a "positive allosteric modulator of nAChR"...
